A comparative expression analysis of gene transcripts in brain tissue of non-transgenic and GH-transgenic zebrafish (Danio rerio) using a DDRT-PCR approach Anais da ABC (AABC)

The presence of higher level of exogenous growth hormone (GH) in transgenic animals could lead to several physiological alterations. A GH transgenic zebrafish (Danio rerio) line was compared to nontransgenic (NT) samples of the species through a DDRT-PCR approach, with the goal of identifying candidate differentially expressed transcripts in brain tissues that could be involved in GH overexpression. Densitometric analyses of two selected amplification products, p300 and ADCY2, pointed to a significant lower gene expression in the transgenic zebrafish (104.02 ± 57.71; 224.10 ± 91.73) when compared to NT samples (249.75 ± 30.08; 342.95 ± 65.19). Gene silencing, also called RNA interference (RNAi) is a specific mechanism of RNA degradation involved in gene regulation, development and defense in eukaryotic organisms. It became an important subject in the teaching programs of molecular biology, genetics and biotechnology courses in the last years. The aim of this work is to provide simple and inexpensive assays to understand and teach gene silencing using plants as model systems. The use of transient and permanent transgenic plants for expressing reporter genes, like those derived from jellyfish green fluorescent protein (gfp) encoding gene, provides a nice, colorful and conclusive image of gene silencing. Apple (Malus domestica Borkh.) is one of the most consumed fruit in the world. Genetic transformation is a key process to sustain this demand by permitting the potential enhancement of existing cultivars as well as the development of new cultivars resistant to pests, diseases, and storage problems that occur in the major production areas. This review summarizes the advances of genetic engineering applied to the development of resistant apple cultivars to fungus disease, with particular attention in the generation of apples resistant to Venturia inaequalis (Cooke) G.Winter, the main phytosanitary problem that affects apple crops in Chile.

An efficient method for constructing transgenic lettuce cultivars by Agrobacterium tumefaciens was described by Torres et al., 1993. In the present work, an improvement of the above procedure is described and applied to transform the cultivar Grand Rapids with a mutated P5CS gene. The major modifications were concerned with turning more practical the transformation and regeneration protocols. Also we tried to improve transformation steps by increasing injured area in explants and prolonging co-cultivation with Agrobacteria (in larger concentration). Genotype-by-environment (GxE) interactions are non-parallel reaction norms among individuals with different genotypes in response to different environmental conditions. GxE interactions are an extension of phenotypic plasticity and consequently studying such interactions improves our ability to predict effects of different environments on phenotype as well as the fitness of genetically distinct organisms and their capacity to interact with ecosystems. This study investigated the impact of constitutively expressed Trichoderma atroviride genes encoding exochitinase nag70 or endochitinase ech42 in transgenic lines of the apple cultivar Pinova on the symbiosis with arbuscular mycorrhizal fungi (AMF). We compared the exo- and endochitinase activities of leaves and roots from non-transgenic Pinova and the transgenic lines T386 and T389. Effects of high proline accumulation on chloroplast and mitochondrial ultrastructure and on osmotic adjustment in tobacco plants Agronomy

Proline accumulates in many organisms in response to abiotic stresses, such as drought and salinity. Reports have discussed the role of proline in relation to osmotic adjustment, even though no clear-cut evidence has yet been provided demonstrating this association as a defense mechanism against drought stress in plants. However, it has been indicated that the application of exogenous proline can damage the ultrastructure of chloroplasts and mitochondria. The initiation and development of organogenic buds and somatic embryos in HBsAg-transgenic cherry tomato (Lycopersicum esculentum var. cerasiforme) mutant were studied histologically. The leaf explants of the mutant were cultured on Murashige & Skoog (MS) basal medium supplemented with 6-BA 1.0 mg/L and IAA 0.05 mg/L for callus induction. Histological studies on the leaf explants of the mutant at various developmental stages revealed that organogenic buds first appeared in the axillary position of explants on the 14th cultured day, and then somatic embryos formed in the same mutant explants after 35 days of culture.
